摘要:
目的 采用代谢组学研究地榆鞣质升高骨髓抑制小鼠白细胞的作用机制。方法 SPF级KM小鼠随机分为正常组、模型组、地榆鞣质组,每组10只。腹腔注射环磷酰胺100 mg/kg诱导小鼠建立骨髓抑制模型,地榆鞣质组按10 mg/kg剂量灌胃给予地榆鞣质水溶液,连续5 d。检测各实验组小鼠外周血液中白细胞数量,并采用LC-MS分析各试验组小鼠血清代谢谱的变化。结果 与模型组比较,地榆鞣质组小鼠白细胞数量显著增加(P<0.05); 经血清代谢轮廓分析,与模型组比较,地榆鞣质组小鼠血清中阿糖胞苷、D-葡糖糖-6磷酸、α-酮异戊酸、十二烷二酸等潜在代谢标志物显著回调,其涉及磷酸戊糖途径、淀粉和蔗糖代谢、异亮氨酸和缬氨酸亮氨酸代谢、半乳糖代谢、糖酵解和糖异生、氨基酸和核苷酸代谢6条代谢路径。结论 地榆鞣质升高骨髓抑制小鼠白细胞可能与调控磷酸戊糖途径,氨基酸和核苷酸代谢的途径等有关。
Abstract:
ObjectiveTo study the mechanisms underlying Sanguisorba tannins increased peripheral blood leukocytes on myelosuppression in mice with metabolomics.Methods The SPF-grade KM mice were randomly divided into: blank group,model group and Sanguisorba tannins group,with 10 mice in each group.Bone marrow suppression model was established in mice induced by intraperitoneal injection of cyclophosphamide(100 mg/kg),and tannin solution was given to the tannin group by gavage at the dose of 10 mg/kg for 5 consecutive days.On the 6th day of the experiment,the counts of peripheral blood leukocytes of the mice in each experimental group was measured,and the changes of serum metabolic profiles of the mice in each experimental group were analyzed by LC-MS.Results Compared with the model group,Sanguisorba tannins significantly increased the counts of peripheral blood leukocytes,elevated potential metabolic markers,such as cytarabine,D-glucose-6 phosphate,α-ketoisovalerate and dodecanedioic acid.By the analysis of serum metabolic profiles,six metabolic pathways of pentose phosphate pathway,starch and sucrose metabolism,isoleucine and valine leucine,galactose metabolism,glycolysis and gluconeogenesis,amino acid and nucleotide metabolism were involved.Conclusion Sanguisorba tannins-increased leukocytes on myelosuppressive in mice might be related to the regulation of pentose phosphate pathway,amino acid and nucleotide metabolism.
