摘要:
目的 探讨杉木提取物凝胶在大鼠皮肤创面愈合过程中的效果。方法 将32只SD大鼠随机分为阴性对照组、阳性对照组、低剂量组及高剂量组,每组8只,将各组大鼠进行全层皮肤缺损造模后,阴性对照组皮肤破损处不予处理,阳性对照组予以莫匹罗星0.1g,低剂量组予以0.1 g/mL杉木凝胶剂0.1 g,高剂量组予以0.2 g/mL杉木凝胶剂0.1g,所有小组大鼠均持续治疗14 d,统计伤后第3 、7 、14 天皮肤愈合率,治疗14d后对愈合处皮肤进行全层取材, HE染色观察皮肤组织愈合情况,采用抑菌试验方法对金黄色葡萄球菌、大肠埃希菌进行杉木原液抗菌效果评估。结果 伤后第3天,高剂量组愈合率高于阴性对照组,低于阳性对照组,低剂量组愈合率低于阳性对照组(P<0.05); 伤后第7天高剂量组愈合率高于阴性对照组(P<0.05),低剂量组愈合率低于阳性对照组(P<0.05); 伤后第14天,高剂量组愈合率高于阴性对照组(P<0.05),低剂量组愈合率明显低于阳性对照组(P<0.05),高于阴性对照组(P<0.05); 与伤前比较,各组伤后第3、7、14天愈合率均显著增高,差异有统计学意义(P<0.05)。与阴性对照组相比,高剂量组与阳性对照组表皮层与真皮层结构清楚、显著增厚、分层明显,且毛细血管增生明显。高剂量与低剂量杉木在金黄色葡萄球菌平板中抑菌圈直径大于生理盐水(P<0.05),小于莫匹罗星(P<0.05); 高剂量杉木在大肠埃希菌平板中抑菌圈直径大于生理盐水(P<0.05),小于莫匹罗星(P<0.05),低剂量杉木在大肠埃希菌平板中抑菌圈直径小于莫匹罗星(P<0.05),与生理盐水无显著差异(P>0.05)。结论 杉木提取物凝胶剂可调控机体炎症反应,促进大鼠皮肤创面的修复,同时具有明显抑菌作用,避免创面感染,利于皮肤伤口愈合。
Abstract:
ObjectiveTo explore the effect of Cunninghamialanceolate extract gel on the healing process of rat skin wounds.Methods Thirty-two SD rats were randomly divided into negative control group,positive control group,low and high doses of Cunninghamialanceolate extract gel groups with 8 rats in each group.After the whole group of skin defects was established,the skin of the negative control group was not treated.The positive control group was given mupirocin(0.1g),the low and high doses of Cunninghamialanceolate extract gel groups were 0.1 g/ml and 0.2g/ml Cunninghamialanceolate gel,respectively.The antibacterial effect of the cedar stock solution on Staphylococcus aureus and Escherichia coli was evaluated by the bacteriostatic test method.Results On the 3rd day after injury,the healing rate of the high dose of Cunninghamialanceolate extract gel group was higher than that in the negative control group.The healing rate of the low dose of Cunninghamialanceolate extract gel group was lower than that in the positive control group.The high rate of the high dose of Cunninghamialanceolate extract gel group was higher on the 7th day after injury.In the negative control group,the healing rate of the low dose of Cunninghamialanceolate extract gel group was lower than that in the positive control group.At the 14th day after injury,the healing rate of the high dose of Cunninghamialanceolate extract gel group was higher than that in the negative control group.Compared with the healing rate before injury,the healing rate in each group was significantly increased on the 3rd,7th and 14th day after injury.Compared with the negative control group,the structure of the epidermis and dermis of the high-dose of Cunninghamialanceolate extract gel group and the positive control group was clear,significantly thickened,and divided.The layer was obvious and the capillary proliferation was obvious.The diameter of inhibition zone of high-dose and low-dose Cunninghamialanceolate in Staphylococcus aureus was larger than that in normal saline,which was smaller than that in mupirocin.The diameter of inhibition zone of high-dose Cunninghamialanceolate in E.coli was larger than that in Normal saline and less than mupirocin.The diameter of the inhibition zone of low-dose Cunninghamialanceolate in E.coli was smaller than that in mupirocin.Conclusion The Cunninghamialanceolate extract gel could regulate the body inflammatory reaction,promote the repair of rat skin wounds,and has obvious antibacterial effect,avoid wound infection and promote skin wound healing.
