摘要:
目的 通过野百合碱(MCT)诱导大鼠肺动脉高压模型研究肺动脉高压发生过程中IL-6、GATA-6在肺组织中的表达及意义。方法 将54只成年雄性SD大鼠随机分成生理盐水对照组(A组,n=18)、空白对照组(B组,n=18)和MCT组(C组,n=18只),C组大鼠给予1%MCT按60mg/kg腹腔内注射造模,A组给予等量0.9%NaCl,B组不做任何处理。分别于造模后第7天(T1)、第14天(T2)、第21天(T3)三个时间点对每组各6只大鼠做右心室压力、右心室肥厚指数监测,qRT-PCR检测肺组织GATA-6表达水平,Western blot检测肺组织IL-6蛋白表达水平,肺组织HE染色观察肺病理改变。结果 与A组、B组比较C组大鼠在T1~T3右心室平均压力及右心室肥厚指数明显增高(P<0.05)且C组中T1~T3右心室平均压力及右心室肥厚指数呈递增性变化并于T3达到最高峰(P<0.05); 与A组、B组比较C组大鼠IL-6表达在T1~T3明显增高(P<0.05)且C组中IL-6表达于T3明显增高并达到最高峰(P<0.05); GATA-6的表达在T1~T3明显降低(P<0.05)且C组中GATA-6的表达于T3明显降低并达到最小峰(P<0.05)。相关分析结果显示C组中IL-6与GATA-6呈显著负相关(r=-0.887,P<0.05)。结论 在肺动脉高压大鼠肺组织中IL-6的表达明显增高,GATA-6的表达明显减低,且两者在肺动脉高压的发生发展过程中可能发挥着重要作用,同时IL-6与GATA-6可能有明显的相关性。
Abstract:
ObjectiveTo investigate the expression and significance of IL-6 and GATA-6 in pulmonary tissues during the occurrence of pulmonary hypertension induced by Monocrotaline(MCT)in rats.Methods A total of 54 adult male SD rats was randomly divided into normal saline control group(group A,n=18),blank control group(group B,n=18),and MCT group(group C,n=18).Rats in group C were given 1% MCT by intraperitoneal injection of 60 mg/kg.At the three time points of day 7(T1),day 14(T2)and day 21(T3)after modeling,formean right ventricular pressure and right ventricular hypertrophy index in each group were monitored.The expression level of GATA-6 in lung tissues was detected by real-time PCR.The expression level of IL-6 protein in lung tissues was detected by western blot assay,and the changes in lung tissues were observed by HE staining.Results Compared with group A and group B,mean right ventricular pressure and right ventricular hypertrophy index in group C were significantly increased.In group C,mean right ventricular pressure and right ventricular hypertrophy index of T1-3 presented increasing changes and reached the peak at T3.Compared with group A and group B,the expression of IL-6 in group C was significantly increased in T1-3,and the expression of IL-6 in T3 was significantly increased and reached the maximum peak in group C.The expression of GATA-6 in group C was significantly decreased in T1-3,and the expression of GATA-6 in T3 was significantly decreased and reached the minimum peak in group C.Conclusion The expression of IL-6 is increased and the expression of GATA-6 is decreased in pulmonary hypertension rat lung tissues.These two proteins may play an important role in the development of pulmonary hypertension,in which IL-6 may be significantly correlated with GATA-6.
