摘要:
目的 探索建立操作简便、高成功率获取大鼠肠淋巴液的方法。方法 采用文献报道的方法和本次实验改进的方法引流大鼠肠淋巴液,对比两组引流成功率、180 min淋巴液引流量以及对淋巴液中包括内毒素(LPS)、肿瘤坏死因子α(TNF-α)、白介素6(IL-6)、白介素4(IL-4)、白介素10(IL-10)等活性物质水平的影响。结果 文献组引流成功率为73.3%(22/30),改进组引流成功率为93.3%(28/30),改进组引流成功率明显较文献组更高(P=0.04),两组大鼠180 min淋巴液引流量及淋巴液中LPS、TNF-α、IL-6、IL-4、IL-10水平比较差异均无统计学意义(P>0.05)。结论 经方法改进后,操作更简便、肠淋巴液引流成功率更高,有利于大鼠肠淋巴液引流模型的建立及后续实验。
Abstract:
ObjectiveTo explore a simple and high success rate method for obtaining rat intestinal lymphatic fluid.Methods The common method reported in the reference and the improved method in this experiment were adopted to drain the intestinal lymphatic fluid of the rats.The success rate of drainage,the drainage volume of lymphatic fluid for 180 min and the effects on the levels of active substances including endotoxin(LPS),tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),interleukin-4(IL-4)and interleukin-10(IL-10)in the two methods groups were compared.Results The success rate of drainage in the reference method group was 73.3%(22/30),the success rate of drainage in the improved method group was 93.3%(28/30).There was no significant difference of lymph drainage in 180 min and the levels of LPS,TNF-α,IL-6,IL-4 and IL-10 between the two methods groups.Conclusion The improved method has simple operation characteristics and higher success rate of lymph drainage,which is conducive to the establishment of lymph drainage model and subsequent experiments.
